ABSTRACT In the chemical and nutritional analyses of pumpkin, the size of bulk sample recommended by the method of reference, makes the process time consuming and difficult to execute. The purpose of the present study was to evaluate different sampling methods to indicate the most suitable one to replace reference method to determine the levels of total carotenoids, total soluble solids, and moisture in pumpkin. A completely randomized design was used with four treatments and 18 replications. Antiproliferative Activity of Violaxanthin Isolated from Bioguided Fractionation of Dunaliella tertiolecta Extracts ArchiMer

Dunaliella tertiolecta (DT) was chemically investigated to isolate molecules inhibiting cancer cell proliferation and inducing apoptosis in vitro. The potency to inhibit cell growth was used for the bio-guided fractionation and isolation of active compounds using chromatographic techniques. The DT dichloromethane extract exhibited a strong anti-proliferative activity on MCF-7 and LNCaP cells, and was further fractionated and sub-fractionated by RP-HPLC. High resolution mass spectrometry and spectrophotometric analysis unequivocally identified violaxanthin as the most antiproliferative molecule present in DT DCM extract. This study was carried out in a plantation of a degraded area in Central Amazon, Brazil, in order to assess environmental light effects on leaf pigment concentrations and chlorophyll a fluorescence in mahogany (Swietenia macrophylla King) and tonka bean (Dipteryx odorata Aubl. Willd). Three-year old saplings were grown in an open site (sun) with maximum radiation of approximately 2000 mumol m-2 s-1 and under the shade of a Ochroma pyramidale (Cav. ex Lam. Urb) canopy. Pfaffia glomerata (Spreng.) Pedersen plantlets were grown under different cadmium (Cd) concentrations (0, 20, 40, 60 and 80 μM) in a hydroponic system during 7 d. Plant growth, micronutrient, chlorophyll and carotenoid concentrations, as well as δ-aminolevulinic acid dehydratase (ALA-D; E.C.4.2.1.24) and acid phosphatase (AP; E.C.3.1.3.2) activities were then analysed. Cadmium concentration in both shoots and roots increased with increasing external Cd levels. Metal concentration was on average 12-fold greater in root than in shoot tissues. Saffron flowers by products including sepals, petals and stamens have no application in Iran’s food industries; nevertheless stigmas contain a considerable amount of carotenoids withhigh antioxidant properties. In this study carotenoid compounds were extracted by an organic solvent (mixture of N-hexane, ethanol and acetone; 2:1:1 v: v) and ultrasound waves. Ultrasound waves were applied at a frequency of 24 kHz and amplitude of 20, 60 and 100% for5, 10 and 15 minute. All treatments were tested  in triplicates. Fruits of Butia capitata (Mart.) Becc as good sources of β -carotene and provitamina Rev. Bras. Frutic.

Butia capitata is a palm tree, widely found in the Brazilian savanna. Their fruits are largely used by local communities to prepare juices, jellies and ice-creams. The main objective of this work was to determine the carotenoids profile of Butia capitata fruits and their provitamin A values. Total carotenoids content ranged from 11.1 to 43.9 μg.g-1; β-carotene was the predominant carotenoid (5.2-22.8 μg.g-1), followed by γ-carotene, phytoene, phytofluene, ζ-carotene, α-cryptoxanthin (or zeinoxanthin) and α-carotene. Provitamin A values varied from 50 to 200 RAE.100g-1. This result suggests that B. capitata pulp may be a good source of β-carotene and provitamin A.

Growth and biopigment accumulation of cyanobacterium Spirulina platensis at different light intensities and temperature BJM

In order to find out optimum culture condition for algal growth, the effect of light irradiance and temperature on growth rate, biomass composition and pigment production of Spirulina platensis were studied in axenic batch cultures. Growth kinetics of cultures showed a wide range of temperature tolerance from 20 ºC to 40 ºC. Maximum growth rate, cell production with maximum accumulation of chlorophyll and phycobilliproteins were found at temperature 35 ºC and 2,000 lux light intensity. But with further increase in temperature and light intensity, reduction in growth rate was observed. Carotenoid content was found maximum at 3,500 lux. Impact of Light Intensity on Antioxidant Activity of Tropical Microalgae ArchiMer

Twelve microalgae species isolated in tropical lagoons of New Caledonia were screened as a new source of antioxidants. Microalgae were cultivated at two light intensities to investigate their influence on antioxidant capacity. To assess antioxidant property of microalgae extracts, four assays with different modes of action were used: 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2’-azinobis (3-éthylbenzothiazoline-6-sulphonique) (ABTS), oxygen radical absorbance capacity (ORAC), and thiobabituric acid reactive substances (TBARS). ABSTRACT: The objective of this research was to evaluate the nutritional composition and bioactive compounds of whole umbu fruit, including pulp, seed and peel, and also of a commercial umbu pulp. Samples of the fractions and of commercial pulp were analyzed for determination of minerals and proximate composition, total phenolic and antioxidant activity. Pulps and peel were also analyzed for vitamin C and carotenoids contents. Commercial pulp presented better nutritional composition than fresh pulp (P<0.05) and the peel presented higher phenolic content and antioxidant activity than seed. Relation between lipid and fatty acid composition of eggs and larval survival in white pacific shrimp (Penaeus vannamei, Boone, 1931) ArchiMer

In this study, lipid classes and fatty acid composition were determined in eggs of P. vannamei as a function of survival to zoea III stage. Spawns were reared individually to zoea III and grouped, according to their final survival to this larval stage, into spawns of high and low survival. Eggs of individual spawns were analyzed for lipid and fatty acid composition of neutral and polar lipids and the results were then grouped according to survival to zoea III. The lipids within each group (high and low survival to zoea III) were pooled for the separation of phospholipids and the analysis of their fatty acid composition. Selection and characterization of carotenoid-producing yeasts from Campinas region, Brazil BJM

The objective of the present study was to select and identify yeasts from Brazil capable of producing carotenoids. Pigmented yeasts were isolated from soil, leaves, fruits, flowers and a processed product. The samples were incubated at 30ºC in Erlenmeyer flasks, containing YM broth. After 48 hours, they were inoculated in Petri dishes with YM agar, and incubated at 30ºC during 120 hours. The yeast colonies, which presented yellow to red coloration, were transferred to culture tubes containing YM agar, and incubated at 30ºC for 72 hours. Torularhodin and torulene are two widespread microbial carotenoids with relatively few studies, as compared to other nutraceutical carotenoids such as β-carotene, lycopene and astaxanthin. Several genera of microorganisms produce it in high concentration (up to 0.1% of the cell dry weight), probably as a protection against photooxidation and free radicals. These pigments, which differ by a terminal carboxylic group, have provitamin-A activity and, being red, have potential use as food and cosmetic color additives. A fast and high-resolution UPLC-MSE analysis was used to identify phytoplankton pigments in an ethanol extract of Porphyridium purpureum (Pp) devoid of phycobiliproteins. In a first step, 22 standard pigments were analyzed by UPLC-MSE to build a database including retention time and accurate masses of parent and fragment ions. Using this database, seven pigments or derivatives previously reported in Pp were unequivocally identified: beta,beta-carotene, chlorophyll a, zeaxanthin, chlorophyllide a, pheophorbide a, pheophytin a, and cryptoxanthin. Zeaxanthin, an abundant carotenoid present in fruits, vegetables and algae was reported to exert antiproliferative activity and induce apoptosis in human uveal melanoma cells. It also inhibited uveal melanoma tumor growth and cell migration in nude mice xenograft models. Here we report that zeaxanthin purified from the rhodophyte Porphyridium purpureum (Bory) K.M.Drew & R.Ross, Porphyridiaceae, promotes apoptosis in the A2058 human melanoma cell line expressing the oncogenic BRAF V600E mutation.
